چکیده

In order to develop a high-efficiency and reproducible transformation protocol for Stylosanthes guianensis we assessed the biological and physical parameters affecting plant electroporation protoplasts. Energy input, as combinations of electric field strengths discharged by different capacitors, electroporation buffer and DNA form were evaluated. Transformation efficiency was assayed in vivo as transient reporter gene expression, using the GFP-coding gene mgfp5 driven by a CaMV 35S constitutive promoter. Energy input and electric field strength had a critical influence on transgene expression with higher transformation levels being achieved with 250 V.cm discharged by 900 and 1000 μF capacitors. Linear plasmid DNA, the absence of chloride and the presence of calcium ions also increased transient gene expression, albeit not significantly.

P-151: Optimal Condition of IntroducingGreen Fluorescent Protein (GFP) Geneinto The Rainbow Trout (Oncorhynchusmykiss) Sperm by Electroporation Method

Background: Many techniques have been developed to increase DNA molecules into sperms called sperm mediated gene transfer (SMGT) and one of them is electroporation. Sperm mediated gene transfer is an ideal method for mass gene transfer in organisms with external fertilization. Gene transfer is applied to rainbow trout sperm and the effects of electroporation on sperms and rate of gene uptake by...

Polymorphism analysis in identification of genetic variation and relationships among Stylosanthes species

A total of 148 accessions representing six important species of the genus Stylosanthes, including S. guianensis, S. hamata, S. scabra, S. seabrana, S. macrocephala, and S. capitata, were used to evaluate genetic variation and relationships using sequence-related amplified polymorphism markers. The results showed that the 18 selected primer pairs generated 138 distinct fragments. The fragment si...

Cloning of Insertion Site Flanking Sequence and Construction of Transfer DNA Insert Mutant Library in Stylosanthes Colletotrichum

Stylosanthes sp. is the most important forage legume in tropical areas worldwide. Stylosanthes anthracnose, which is mainly caused by Colletotrichum gloeosporioides, is a globally severe disease in stylo production. Little progress has been made in anthracnose molecular pathogenesis research. In this study, Agrobacterium tumefaciens-mediated transformation was used to transform Stylosanthes col...

Electroporated intact BY-2 tobacco culture cells as a model of transient expression study.

Transfer of foreign genes into plant cells can be accomplished by several methods: agrobacterium-mediated, microinjection, biolistic particle bombardment and electroporation. The last one is frequently used for transfection of plant protoplasts for transient gene expression. Electroporation is a simple procedure and allows transfecting a large number of cells at one time. Square wave-modulated ...

Expression of genes transferred into monocot and dicot plant cells by electroporation.

We have developed a general method for electrically introducing DNA into plant cells. Gene transfer occurs when a high-voltage electric pulse is applied to a solution containing protoplasts and DNA. Carrot protoplasts were used as a model system to optimize gene-transfer efficiency, which was measured 24-48 hr after electroporation by the amount of chloramphenicol acetyltransferase activity res...
